USE OF n - ALKANE AND LIPID FORMATION BY A NOCARDIA
نویسنده
چکیده
Utilization of various hydrocarbons by microbes is well known (Beerstecher, 1954), but little has been reported on the cellular composition of hydrocarbongrown cells. No doubt it has been assumed that these cells differ little from those cultivated on nonhydrocarbon substrates and, in general, this assumption is probably true. But it is recognized that substrates and cultural conditions can influence not only extracellular products but also the composition of microbial cells. In the course of studying conditions for rapid utilization of hydrocarbons by microbes we have observed a marked increase in the lipid content of nocardia grown on liquid n-alkanes. This was noted particularly in the case of n-hexadecane and n-octadecane cultures. As much as 70 per cent of the cellular material formed from these substrates can be extracted with fat solvents. This lipid consists principally of glycerides and straight-chain waxes. In contrast, nocardial cells grown on n-hexane or n-tridecane contain about the same amount of lipid as glucose-grown cells, less than 30 per cent, and no waxes. This paper describes the conditions used for cultivating a soil-isolated nocardia on liquid n-alkanes; the conversion of n-alkane to gross cellular products; and some of the factors which influence growth and cellular lipid formation.

متن کاملMicrobial Incorporation of Fatty Acids Derived from N-alkanes into Glycerides and Waxes.
When n-alkanes with 13 to 20 carbon atoms were fed to a Nocardia closely related to N. salmonicolor, the produced cellular triglycerides and aliphatic waxes invariably contained fatty acids with an even or an odd number of carbon atoms subject to this feature of the n-alkane substrate. Beta-oxidation and C(2) addition are both operative, as evidenced by the spectra of fatty acids incorporated i...
متن کاملFormation of planar bilayer membranes from lipid monolayers. A critique.
The formation of planar bilayer membranes from lipid monolayers as described by Montal and Mueller (Proc. Natl. Acad. Sci. 1972. 69:3561) is analyzed. Bilayers absolutely free of alkane solvents or other nonpolar hydrocarbons can be formed on polytetrafluoroethylene (PTFE) (e.g. Teflon) septa only if certain boundary conditions are satisfied. Measurements have been made of the contact angles be...
متن کاملCharacterization and two-dimensional crystallization of membrane component AlkB of the medium-chain alkane hydroxylase system from Pseudomonas putida GPo1.
The alkane hydroxylase system of Pseudomonas putida GPo1 allows it to use alkanes as the sole source of carbon and energy. Bacterial alkane hydroxylases have tremendous potential as biocatalysts for the stereo- and regioselective transformation of a wide range of chemically inert unreactive alkanes into valuable reactive chemical precursors. We have produced and characterized the first 2-dimens...
متن کاملEnhancing Alkane Production in Cyanobacterial Lipid Droplets: A ModeFl Platform for Industrially Relevant Compound Production
Cyanobacterial lipid droplets (LDs) are packed with hydrophobic energy-dense compounds and have great potential for biotechnological expression and the compartmentalization of high value compounds. Nostoc punctiforme normally accumulates LDs containing neutral lipids, and small amounts of heptadecane, during the stationary phase of growth. In this study, we further enhanced heptadecane producti...
